Biuret reagent is normally blue but it changes colour to violet in the presence of protein because Cu²⁺ of the reagent binds to the peptide bonds of protein. So, a mixture of protein and Biuret reagent is violet. However, when proteins are heated, they denature, which means their peptide bonds are broken down. Also, the complex between Biuret reagent and now destroyed peptide bonds does not exist any more, the reagent is free again and therefore, blue.

The color change is as a result of the break in peptide bonds in the protein as a result of denaturation by heat.

What is Denaturation?

This is the process in which a compound deviates from its original state  due to the loss of structures.

Biuret reagent has a blue color but changes to violet in the presence of protein. When proteins are heated, they are denatured and the peptide bonds does not exist any more which is why the the reagent changes back to blue.
